Environmental xenoestrogens pose a significant health risk for all living organisms. There is growing evidence concerning the different susceptibility to xenoestrogens of developing and adult organisms, but little is known about their genotoxicity in pre-pubertal mammals. In the present study, we developed an animal model to test the sex- and age-specific genotoxicity of the synthetic estrogen diethylstilbestrol (DES) on the reticulocytes of 3-week-old pre-pubertal and 12-week-old adult BALB/CJ mice using the in vivo micronucleus (MN) assay. The aim of this study was to evaluate the therapeutic and toxicologic effects of the administration of the powered vegetable extract and aqueous extract of Tournefortia paniculata leaves on Wistar rats, subjected to a hypercaloric diet for 42 days. The rats were divided into five groups and were given the following treatments by gavage: T0 (control) - 1.0 mL water day-1; T1 - aqueous extract containing 14 mg phenolic compounds kg-1 rat day-1; T2 - 14 mg quercetin kg-1 rat day-1; T3 - 50 mg powered vegetable extract from T. paniculata leaves (PVE) kg-1 rat day-1 and T4 - 100 mg PVE kg-1 rat day-1. This study was conducted to evaluate the potential induction of genotoxic effects of white bean flour using the Comet assay. The test was conducted with human lymphocytes present in whole blood immediately after collection, by incubation with white bean flour in three concentrations (3.92, 9.52 and 18.18 mg/mL) at 37 ºC for 4 h followed by preparation of slides. Samples were considered positive (above 20% damage) when the damage observed to cellular DNA was higher than the negative control. No genotoxic potential was found at the doses tested. Duguetia furfuracea (St. Hil.) Benth & Hook f. (1862), popularly known as "sofre-do-rim-quem-quer" and "araticum-seco", is a shrub of the Annonaceae family that occurs in several regions of Brazil. In folk medicine the infusion of its leaves and twigs is used to treat rheumatism and renal colic, whereas the seed powder is mixed with water to treat pediculosis. In the present study, the ability of Punica granatum ethanolic leaf extract (PGL) and Punica granatum ethanolic fruit extract (PGF) to induce mutagenicity or to modulate the genotoxic effects induced by the alkylating agent cyclophosphamide (CP) was evaluated. Swiss male mice were treated by gavage for 10 days with PGL or PGF (12.5, 25, 50, and 75 mg/kg/day) prior to exposure to CP (i.p. 200 mg/kg), 24 h after the end of the treatment. Resumo: Dentre a variedade de bioensaios disponíveis para estudos de detecção e monitoração dos efeitos tóxicos genéticos de substâncias e ambientes contaminados, os bioensaios que empregam a planta Tradescantia destacam-se pela sensibilidade e facilidade de realização de estudos de curto prazo in situ. Este trabalho apresenta uma revisão extensiva dos estudos de mutagênese ambiental empregando Tradescantia como bioindicador, e é complementado com resultados experimentais e indicações metodológicas.

This study assessed the limnology from the Medium Uruguay River Basin in Uruguaiana, Brazil, with a focus on the concentration of heavy metals (Cd, Cu, and Mn), to assess the toxicological potential (cytotoxicity and genotoxicity) for humans using as biological matrix of study human leukocyte cells. The conductivity, resistivity, and dissolved O2 levels exceeded the limits recommended by the National Environmental Council (Conselho Nacional do Meio Ambiente - CONAMA). Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous contaminants that can be present at high levels as mixtures in polluted aquatic environments. Many PAHs are potent mutagens and several are well-known carcinogens. Despite numerous studies on individual compounds, little is known about the toxicity of PAHs mixtures that are encountered in environmental situations. In the present work, zebrafish were continuously fed from 5 days post-fertilisation to 14 months post-fertilisation (mpf) with a diet spiked with fractions of either pyrolytic (PY), petrogenic light oil (LO), or petrogenic heavy oil (HO) origin at three concentrations. Although alcohol is known to be a carcinogen for humans, ethanol-genotoxicity studies are incomplete. Ethanol seems not to be a bacterial mutagen, but the results are conflicting in rodent assays. We investigate the genotoxicity in the bone marrow micronucleus (MN) test and in the dominant lethal mutation (DLM) assay using two long-term ethanol exposure protocols. In the MN test, mice consumed three doses (5, 10 and 15% v/v) for 32 weeks. The alkaline comet assay was tested on different microalgae: the clinofiagellates, Karenia mikimotoi and Alexandrium minuturn, and the diatom, Chaetoceros gracilis. The microalgae were exposed during their exponential growth to the model direct genotoxicant, hydrogen peroxide (1 h, 5 and 100 pM H2O2). Following H2O2 exposure, the comet assay was validated only for K. mikimotoi for which genotoxicity was observed from the lowest tested concentration of 5 pM with a concentration-dependent effect. In this work, we used the comet assay to examine the genotoxicity of glyphosate isopropylamine (0.7, 7, 70 and 700 µM) in human lymphocytes, erythrocytes of Oreochromis niloticus and staminal nuclei of Tradescantia (4430) in vitro and in vivo. Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG

The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. In this study, the onion (Allium cepa) root test was used to evaluate the genotoxicity of the Pitimbu River (Natal city, Brazil) surface water. The water was collected at five sampling sites along the river and one sample was obtained after the treatment (flocculation, chlorination and pH correction) of the river water for human consumption. All raw river water samples increased the frequency of chromosomal abnormalities and/or micronuclei and two of the water samples produced alterations in the mitotic index of the root cells. Two of the water samples also altered root growth and two produced morphological modifications in the A. cepa roots. Mercury is a xenobiotic metal that is a highly deleterious environmental pollutant. The biotransformation of mercury chloride (HgCl2) into methylmercury chloride (CH3HgCl) in aquatic environments is well-known and humans are exposed by consumption of contaminated fish, shellfish and algae. The objective of the present study was to determine the changes induced in vitro by two mercury compounds (HgCl2 and CH3HgCl) in cultured human lymphocytes. Short-term human leukocyte cultures from 10 healthy donors (5 females and 5 males) were set-up by adding drops of whole blood in complete medium.
